Nanoscale chemical mapping of newly-synthesised phospholipid molecules inside a mammlian cell is demonstrated using tip-enhanced Raman spectroscopy (TERS) for the first time. Mouse pre-adipocyte cells pulse-treated with deuterated sodium stearate, are used as a model system to showcase TERS mapping. Newly-synthesised membrane phospholipid distribution is mapped with less than 20 nm spatial resolution by overcoming the diffraction limit of confocal Raman spectroscopy using strong plasmonic enhancement of Raman signals at a TERS tip-apex.
